Barley (Hordeum vulgare L.) is a cereal grass mainly used as animal fodder and raw material for the malting industry. The map-based reference genome sequence of barley cv. 'Morex' was constructed by the International Barley Genome Sequencing Consortium (IBSC) using hierarchical shotgun sequencing. Here, we report the experimental and computational procedures to (i) sequence and assemble more than 80,000 bacterial artificial chromosome (BAC) clones along the minimum tiling path of a genome-wide physical map, (ii) find and validate overlaps between adjacent BACs, (iii) construct 4,265 non-redundant sequence scaffolds representing clusters of overlapping BACs, and (iv) order and orient these BAC clusters along the seven barley chromosomes using positional information provided by dense genetic maps, an optical map and chromosome conformation capture sequencing (Hi-C). Integrative access to these sequence and mapping resources is provided by the barley genome explorer (BARLEX).

A forward genetic screen identified a dominant-negative Arabidopsis thaliana mutant resistant to growth inhibition caused by the actin-disrupting compound latrunculin B (LatB). Map-based cloning and whole-genome sequencing revealed that the mutant referred to here as lbr1 for LatB resistant1 had a point mutation in the AT5G09810 gene, which encodes the vegetative actin (ACT) isoform ACT7. The cytosine to thymine mutation in exon two of lbr1 ACT7 led to substitution of proline to serine at position 34 (P34S) adjacent to the nucleotide-binding cleft of the ACT7 protein.

It threatens food and nutrition security, depriving low-income consumers' access to healthy, affordable food and quality nutrition.

Spatial perspective taking (SPT) is a core cognitive ability essential for real-world navigation, yet the examination of dynamic integration of allocentric and egocentric reference frames using map has received limited attention. To address this gap, the present study introduces a Map-Based Self-Localization Task paired with event-related potential (ERP) techniques to examine how angular disparities and rotation directions affect SPT. High school participants (n = 38) completed 320 Map-Based Self-Localization trials.

, an essential plant in traditional Chinese medicine, encounters significant challenges in the development of germplasm resources and cytological research. This study employed the reference genome to develop 20 repetitive sequence oligonucleotide (oligo) probes, all of which produced clear signals on the chromosomes of the cultivar Qicun Xiang Ai (QCXA). These probes were configured into two probe cocktails (Multiplex #1 and Multiplex #2) that effectively generated chromosome signals under non-denaturing hybridization conditions through probe staining.

Duck is one of the most widely distributed waterfowl in the world, with more than 6 billion of them farmed annually in the world, and has great economic and ecological value. Amidst mounting global prioritization of duck genetic resource exploration and prevalent inter-varietal hybridization events, the traditional breed identification methods are difficult to address actual requirements, restricting the utilization, development and protection of duck germplasm resources. This study aims to develop an accurate, efficient, and scalable duck DNA fingerprinting system based on genomic technologies and machine learning methods to address the urgent need for breed identification tools in high-quality agricultural production and ecological protection.
