Oligomers of the 42-mer amyloid-β protein (Aβ42), rather than fibrils, cause synaptic dysfunction in the pathology of Alzheimer's disease (AD). The nucleation phase in a nucleation-dependent aggregation model of Aβ42 is related to the formation of oligomers. Uncaria rhynchophylla is one component of "Yokukansan", a Kampo medicine, which is widely used for treating AD symptoms. Previously, an extract of U. rhynchophylla was found to reduce the aggregation of Aβ42, but its active principles have yet to be identified. In the present work, uncarinic acid C (3) was identified as an inhibitor of Aβ42 aggregation that is present in U. rhynchophylla. Moreover, compound 3 acted as a specific inhibitor of the nucleation phase of Aβ42 aggregation. Compound 3 was synthesized from saponin A (10), an abundant byproduct of rutin purified from Uncaria elliptica. Comprehensive structure-activity studies on 3 suggest that both a C-27 ferulate and a C-28 carboxylic acid group are required for its inhibitory activity. These findings may aid the development of oligomer-specific inhibitors for AD therapy.

The binary composites of liquid () and crystalline () difluoroboron β-diketonate (BFdbk) complexes exhibited a metastable nature arising from the intricate interplay between their liquid and crystalline components in bulk. Differential scanning calorimetry (DSC) measurements indicate nearly complete miscibility of and when the fractional volume of occupied a substantial portion, corresponding to below 47 mol % of the content. In contrast, polarized optical microscopic (POM) observations unveiled that the / composites between two glass slides crystallized regardless of the content.

Joint direct microscopy-calorimetry measurements of crystal growth were performed for a 60 nm amorphous Sb2S3 film deposited either on a Kapton foil or on a soda-lime glass. Calorimetric crystallization proceeded in two steps, originating either from mechanical and stress-induced defects (230-275 °C) or from homogeneously formed nuclei (255-310 °C); both processes exhibited an identical activation energy of 200 kJ mol-1. At temperatures <230 °C, a Sb2O3 crystalline phase formed along the rhombohedral Sb2S3 structure.

The equilibration dynamics of ultrastable glasses subjected to heating protocols has attracted recent experimental and theoretical interest. With simulations of the mW water model, we investigate the devitrification and "melting" dynamics of both conventional quenched (QG) and vapor deposited (DG) amorphous ices under controlled heating ramps. By developing an algorithm to reconstruct hydrogen-bond networks, we show that bond ring statistics correlate with the structural stability of the glasses and allow tracking crystalline and liquid clusters during devitrification and melting.
