Alternative splicing (AS) defects that adversely affect gene expression and function have been identified in diabetic hearts; however, the mechanisms responsible are largely unknown. Here, we show that the RNA-binding protein RBFOX2 contributes to transcriptome changes under diabetic conditions. RBFOX2 controls AS of genes with important roles in heart function relevant to diabetic cardiomyopathy. RBFOX2 protein levels are elevated in diabetic hearts despite low RBFOX2 AS activity. A dominant-negative (DN) isoform of RBFOX2 that blocks RBFOX2-mediated AS is generated in diabetic hearts. DN RBFOX2 interacts with wild-type (WT) RBFOX2, and ectopic expression of DN RBFOX2 inhibits AS of RBFOX2 targets. Notably, DN RBFOX2 expression is specific to diabetes and occurs at early stages before cardiomyopathy symptoms appear. Importantly, DN RBFOX2 expression impairs intracellular calcium release in cardiomyocytes. Our results demonstrate that RBFOX2 dysregulation by DN RBFOX2 is an early pathogenic event in diabetic hearts.

Pompe disease is a glycogen storage disorder caused by mutations in the acid α-glucosidase (GAA) gene, leading to reduced GAA activity and glycogen accumulation in heart and skeletal muscles. Enzyme replacement therapy with recombinant GAA, the standard of care for Pompe disease, is limited by poor skeletal muscle distribution and immune responses after repeated administrations. The expression of GAA in muscle with adeno-associated virus (AAV) vectors has shown limitations, mainly the low targeting efficiency and immune responses to the transgene.

Background: The objective of this study was to compare the effects of daily consumption of white potatoes compared with white rice on cardiometabolic health in individuals with type-2 diabetes (T2D).
Objective: To determine the effects of white potato consumption compared to white rice (a commonly consumed refined grain) on indices of glycemic control and cardiovascular health in individuals with overweight or obesity and T2D.
Methods: In this randomized crossover study, comparative control trial, 24 adults with T2D [45-80 y, body mass index (kg/m) 25-40] consumed baked white potatoes (100 g) or calorie-matched white rice (75 g) daily for 12 wk, separated by a 2-wk washout, with assessments of glycemic control, lipids, inflammation, blood pressure, endothelial function, and body composition at baseline (only 1 baseline visit included as a covariate in statistical analyses), 6 wk, and 12 wk.

Endovascular mechanical thrombectomy (MT) is a recommended treatment for acute ischemic stroke due to large vessel occlusion (LVO). The objective of the present study was to evaluate the impact of vascular risk factors on the outcome of MT outcomes in patients with stroke with LVO and to determine the prevalence of structural epilepsy in these patients. Background: While psychological stress cannot be dissociated from chronic diseases, the extent to which it impacts the management of chronic diseases is poorly understood. This cross-sectional study investigated the prevalence and impact of psychological stress among Rwandan patients with chronic diseases, particularly hypertension, heart failure, malignancies, diabetes, and kidney failure.
Methods: This cross-sectional study was conducted among internal medicine patients receiving treatment for chronic diseases at the University Teaching Hospital of Kigali (n = 81) and the University Teaching Hospital of Butare (CHUB) (n = 78) between May 1 and June 30, 2024.

Metabolic dysfunction-associated steatotic liver disease (MASLD) has emerged as a major contributor to systemic metabolic dysfunction and is increasingly recognized as a risk enhancer for both cardiovascular disease (CVD) and chronic kidney disease (CKD). This review explores the complex interconnections between MASLD, CVD, and CKD, with emphasis on shared pathophysiological mechanisms and the clinical implications for risk assessment and management. We describe the crosstalk among the liver, heart, and kidneys, focusing on insulin resistance, chronic inflammation, and progressive fibrosis as key mediators.
