The mixed lineage kinase ZAK is a key regulator of the MAPK pathway mediating cell survival and inflammatory response. ZAK is targeted by several clinically approved kinase inhibitors, and inhibition of ZAK has been reported to protect from doxorubicin-induced cardiomyopathy. On the other hand, unintended targeting of ZAK has been linked to severe adverse effects such as the development of cutaneous squamous cell carcinoma. Therefore, both specific inhibitors of ZAK, as well as anticancer drugs lacking off-target activity against ZAK, may provide therapeutic benefit. Here, we report the first crystal structure of ZAK in complex with the B-RAF inhibitor vemurafenib. The cocrystal structure displayed a number of ZAK-specific features including a highly distorted P loop conformation enabling rational inhibitor design. Positional scanning peptide library analysis revealed a unique substrate specificity of the ZAK kinase including unprecedented preferences for histidine residues at positions -1 and +2 relative to the phosphoacceptor site. In addition, we screened a library of clinical kinase inhibitors identifying several inhibitors that potently inhibit ZAK, demonstrating that this kinase is commonly mistargeted by currently used anticancer drugs.

Background: Bariatric surgery pre-operative workup mandates many multidisciplinary visits demanding patient's commitment in time and travel. Due to the COVID pandemic, our bariatric clinic transitioned to a telemedicine model. The objective of this work is to determine the impact of this shift.

The objective of this study was to develop a population pharmacokinetic model for linezolid in hematooncological patients with sepsis, and to propose dosing optimization based on pharmacokinetic covariates that would lead to improved achievement of the PK/PD target. Therapeutic drug monitoring data from hematooncological patients treated with linezolid for suspected or proven sepsis were analyzed. A pharmacokinetic population model for linezolid was constructed using a nonlinear mixed-effects modeling approach.

We aimed to evaluate the efficacy and safety of sotrovimab in the RECOVERY trial, an investigator-initiated, individually randomised, controlled, open-label, adaptive platform trial testing treatments for patients admitted to hospital with COVID-19.
Methods: Patients admitted with COVID-19 pneumonia to 107 UK hospitals were randomly assigned (1:1) to either usual care alone or usual care plus a single 1 g infusion of sotrovimab, using web-based unstratified randomisation.

In this study, we investigate various geometric aspects of a photonic hexagonal lattice made of triple-leg stripline resonators (TSRs) in a circuit QED system. The inherent two-fold degenerate spatial modes of the TSR act as two distinct orbitals in our 2D lattice system. Remarkably the energy spectra of the system exhibits the dispersive quadratic band-touching to the top and bottom flat bands.

Easily accessible and inexpensive potassium carbonate (KCO) has been applied as the base-catalyst for the synthesis of novel classes of functionalized nanomaterials. This eco-friendly approach has been proven to be effective for a wide range of substrates, leading to nine isocyanate derivatives of silsesquioxanes (SQs) with yields exceeding 90% in mild and transition metal-free conditions. The application potential of chosen products was assessed on the basis of thermogravimetric analyses and photochemical measurements.
