The efficacy of vaccine adjuvants such as Toll-like receptor agonists (TLRa) can be improved through formulation and delivery approaches. Here, we attached small molecule TLR-7/8a to polymer scaffolds (polymer-TLR-7/8a) and evaluated how different physicochemical properties of the TLR-7/8a and polymer carrier influenced the location, magnitude and duration of innate immune activation in vivo. Particle formation by polymer-TLR-7/8a was the most important factor for restricting adjuvant distribution and prolonging activity in draining lymph nodes. The improved pharmacokinetic profile by particulate polymer-TLR-7/8a was also associated with reduced morbidity and enhanced vaccine immunogenicity for inducing antibodies and T cell immunity. We extended these findings to the development of a modular approach in which protein antigens are site-specifically linked to temperature-responsive polymer-TLR-7/8a adjuvants that self-assemble into immunogenic particles at physiologic temperatures in vivo. Our findings provide a chemical and structural basis for optimizing adjuvant design to elicit broad-based antibody and T cell responses with protein antigens.

This study demonstrates the successful fabrication of nanostructured Langmuir-Blodgett (LB) films combining the conjugated copolymer poly(9,9-dioctylfluorene--3,4-ethylenedioxythiophene) (PDOF--PEDOT) with spherical and triangular silver nanoparticles (AgNP). The LB technique allowed precise control over the molecular arrangement and distribution of the nanoparticles at the air-water interface, resulting in compact, reproducible and structurally ordered nanocomposite films. The structural and morphological properties of the interfacial monolayers and LB films were investigated using surface pressure-area isotherms, Brewster angle microscopy, polarization modulation infrared reflection-absorption spectroscopy (PM-IRRAS) and quartz crystal microbalance.

Nimodipine (NMP), a poorly water-soluble small-molecule agent, demonstrates notable therapeutic limitations in addressing cerebral vasospasm secondary to subarachnoid hemorrhage (SAH). Owing to its inherent physicochemical properties characterized by low oral bioavailability, rapid elimination half-life, and extensive first-pass metabolism, conventional formulations necessitate frequent dosing regimens to sustain therapeutic plasma concentrations. These pharmacological challenges collectively result in suboptimal patient adherence, marked plasma concentration fluctuations, and recurrent vascular irritation.

The multifunctional systems presented here introduce an innovative and deeply thought-out approach to the more effective and safer use of temozolomide (TMZ) in treating glioma. The developed hydrogel-based flakes were designed to address the issues of local GBL therapy, bacterial neuroinfections, and the bleeding control needed during tumor resection. The materials obtained comprise TMZ and vancomycin (VANC) loaded into cyclodextrin/polymeric capsules and embedded into gelatin/hyaluronic acid/chitosan-based hydrogel films cross-linked with genipin.

This study integrates machine learning (ML) and density functional theory (DFT) to systematically investigate the oxygen electrocatalytic activity of two-dimensional (2D) TM(HXBHYB) (HX/YB = HIB (hexaaminobenzene), HHB (hexahydroxybenzene), HTB (hexathiolbenzene), and HSB (hexaselenolbenzene)) metal-organic frameworks (MOFs). By coupling transition metals (TM) with the above ligands, stable 2D TM(HXBHYB)@MOF systems were constructed. The Random Forest Regression (RFR) model outperformed the others, revealing the intrinsic relationship between the physicochemical properties of 2D TM(HXBHYB)@MOF and their ORR/OER overpotentials.
