The second messenger molecule cAMP regulates the activation phase of the cAMP signaling pathway through high-affinity interactions with the cytosolic cAMP receptor, the protein kinase A regulatory subunit (PKAR). Phosphodiesterases (PDEs) are enzymes responsible for catalyzing hydrolysis of cAMP to 5' AMP. It was recently shown that PDEs interact with PKAR to initiate the termination phase of the cAMP signaling pathway. While the steps in the activation phase are well understood, steps in the termination pathway are unknown. Specifically, the binding and allosteric networks that regulate the dynamic interplay between PKAR, PDE, and cAMP are unclear. In this study, PKAR and PDE from Dictyostelium discoideum (RD and RegA, respectively) were used as a model system to monitor complex formation in the presence and absence of cAMP. Amide hydrogen/deuterium exchange mass spectrometry was used to monitor slow conformational transitions in RD, using disordered regions as conformational probes. Our results reveal that RD regulates its interactions with cAMP and RegA at distinct loci by undergoing slow conformational transitions between two metastable states. In the presence of cAMP, RD and RegA form a stable ternary complex, while in the absence of cAMP they maintain transient interactions. RegA and cAMP each bind at orthogonal sites on RD with resultant contrasting effects on its dynamics through parallel allosteric relays at multiple important loci. RD thus serves as an integrative node in cAMP termination by coordinating multiple allosteric relays and governing the output signal response.

The small GTPase Rho5 has been shown to be involved in regulating the Baker's yeast response to stress on the cell wall, high medium osmolarity, and reactive oxygen species. These stress conditions trigger a rapid translocation of Rho5 and its dimeric GDP/GTP exchange factor (GEF) to the mitochondrial surface, which was also observed upon glucose starvation. We here show that rho5 deletions affect carbohydrate metabolism both at the transcriptomic and the proteomic level, in addition to cell wall and mitochondrial composition.

Purpose: To evaluate the impact of MRP inhibition by MK571 on prostate hypercontractility in diet-induced obesity, based on the hypothesis that this intervention enhances intracellular cAMP and cGMP signaling.
Methods: Adult C57BL/6 mice were divided into three groups: (i) lean, (ii) obese, and (iii) obese + MK571 (5 mg/kg/day, 14 days). The prostate was isolated for immunohistochemistry, biochemistry and functional assays.

Food combinations featuring specific functional components represent one of the effective intervention strategies for alleviating functional gastrointestinal disorders induced by dietary and environmental factors. Honey and aloe vera have both been recognized as natural agents with laxative effects, yet the synergistic effects of their combination in alleviating constipation and the underlying regulatory mechanism remain to be elucidated. This study formulated a honey-aloe paste by employing honey as the primary ingredient compounded with aloe vera gel and investigated its preventive effects on loperamide-induced slow-transit constipation through a comprehensive analysis of gastrointestinal function and intestinal microenvironment.

We compared the effects of ex-vivo machine perfusion (EVMP) of hearts donated after circulatory death (DCD) with the single-shot solutions HTK-N and Del Nido cardioplegia (DNC) on left-ventricular (LV) contractility and myocardial microcirculation. In a DCD pig model, hearts were maintained by EVMP with hypothermic, oxygenated HTK-N (DCD-HTK-N; N = 8) or DNC (DCD-DNC; N = 8) followed by reperfusion with blood, including assessment of contractility and microcirculation with Laser-Doppler-Flow (LDF). We performed transcriptomics using microarrays.

Introduction: Summer day camps (SDC) can mitigate summer weight gain by providing a structured daily environment that promotes healthy behaviors, but SDCs are often cost prohibitive to families with low-income. This study evaluated the cost effectiveness of providing free SDC to disadvantaged children to prevent summer weight gain.
Methods: 422 children from a low-income school district in South Carolina were recruited and randomly assigned to receive 8-10 weeks of free SDC or to experience summer as usual in 2021-2023.
