Most cell-surface receptors for cytokines and growth factors signal as dimers, but it is unclear whether remodeling receptor dimer topology is a viable strategy to "tune" signaling output. We utilized diabodies (DA) as surrogate ligands in a prototypical dimeric receptor-ligand system, the cytokine Erythropoietin (EPO) and its receptor (EpoR), to dimerize EpoR ectodomains in non-native architectures. Diabody-induced signaling amplitudes varied from full to minimal agonism, and structures of these DA/EpoR complexes differed in EpoR dimer orientation and proximity. Diabodies also elicited biased or differential activation of signaling pathways and gene expression profiles compared to EPO. Non-signaling diabodies inhibited proliferation of erythroid precursors from patients with a myeloproliferative neoplasm due to a constitutively active JAK2V617F mutation. Thus, intracellular oncogenic mutations causing ligand-independent receptor activation can be counteracted by extracellular ligands that re-orient receptors into inactive dimer topologies. This approach has broad applications for tuning signaling output for many dimeric receptor systems.

The crystallographic B-factor (Bf), also known as the Debye-Waller factor (DWF) or temperature factor, relates to the mean-square displacement of the atoms (X). X may be composed of individual contributions from lattice disorder (LT), static conformational heterogeneity (H) throughout the lattice, rigid body vibration (RB), local conformational vibration (V), and zero-point atomic fluctuation (A). The Bf has been widely employed as a surrogate measure of local protein flexibility, although such relation has not been confirmed.

Aluminum nanoparticles (Al NPs) were synthesized via catalyzed thermal reduction of an aluminum precursor in the presence of a capping ligand. A systematic study was conducted to examine the effect of dilution on nanoparticle synthesis by varying the volume of anhydrous toluene across four dilution factors while maintaining constant molar quantities of the aluminum precursor, catalyst, and ligand. This methodology is relevant for scale-up processes, where more dilute conditions can mitigate nanoparticle reactivity and enhance safety.

Background: This study aimed to evaluate the relationship between surrogate efficacy outcomes and overall survival (OS) in clinical trials for recurrent or metastatic head and neck squamous cell carcinoma (R/M HNSCC), and to develop a predictive model for OS that incorporates these surrogate outcomes while accounting for baseline patient characteristics.
Materials And Methods: Data were systematically collected from first-line trials published between January 2010 and March 2025 for R/M HNSCC. Five machine learning models were assessed to predict OS based on surrogate outcomes [objective response rate (ORR), disease control rate, progression free survival (PFS), duration of response, 1-year OS rate] and patient characteristics [human papillomavirus (HPV) status, Eastern Cooperative Oncology Group (ECOG) performance status, programmed death-ligand 1 (PD-L1) expression].

Quantitation of human immunodeficiency virus-1 (HIV-1) broadly neutralizing antibodies (bNAbs) in human serum is required for clinical trials investigating the pharmacokinetics, pharmacodynamics, and drug interactions of these treatments. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) is gaining interest as an alternative to ligand binding for therapeutic antibody quantitation in serum.
