Remote focussing microscopy allows sharp, in-focus images to be acquired at high speed from outside of the focal plane of an objective lens without any agitation of the specimen. However, without careful optical alignment, the advantages of remote focussing microscopy could be compromised by the introduction of depth-dependent scaling artifacts. To achieve an ideal alignment in a point-scanning remote focussing microscope, the lateral (XY) scan mirror pair must be imaged onto the back focal plane of both the reference and imaging objectives, in a telecentric arrangement. However, for many commercial objective lenses, it can be difficult to accurately locate the position of the back focal plane. This paper investigates the impact of this limitation on the fidelity of three-dimensional data sets of living cardiac tissue, specifically the introduction of distortions. These distortions limit the accuracy of sarcomere measurements taken directly from raw volumetric data. The origin of the distortion is first identified through simulation of a remote focussing microscope. Using a novel three-dimensional calibration specimen it was then possible to quantify experimentally the size of the distortion as a function of objective misalignment. Finally, by first approximating and then compensating the distortion in imaging data from whole heart rodent studies, the variance of sarcomere length (SL) measurements was reduced by almost 50%.

The Darwin Mounds marine protected area has been afforded protection from bottom trawl fishing since August 2003, following the discovery of cold-water coral (CWC) communities in June 1998. Surveys of the area in 2000 provided evidence that deep-water trawling activities were impacting the corals, prompting the development of a conservation response. Here we report the most recent survey of these CWCs in 2019, contrasting those data with a prior survey in 2011, and the earliest observations in the area (1998-2000).

Aims: To assess the acceptability and feasibility of an individual-focussed, six-session virtual coaching program and its potential effectiveness on self-reported levels of psychological distress, well-being, flourishing, and solution-focussed thinking in a cohort of Australian senior doctors.
Methods: Program delivery occurred between 1 August 2020 and 8 January 2021.

The delegation of medical tasks to medical assistants (MFA) offers a promising approach to relieve the burden on GPs and to support the provision of sustainable care. The 'VERSORGT am ORT' (VaO) project develops this principle and aims to reduce working hours away from patients by partially shifting delegated home visits to specialised care rooms without compromising the quality of care.

Methods: The research focusses on a pre-implementation phase assessing the knowledge and acceptance levels of XR technologies among healthcare professionals and the general population.
