The aim of this review article is to summarize the information available related to milk fat globules (MFGs) in order to highlight their contribution to the nutritional and technological characteristics of milk and dairy products. The macrostructure of milk lipids is composed of globules made up of triglycerides with different melting points, enveloped by a biological membrane from the mammary epithelial cell. In milks of different animal species, there are different-sized MFGs, ranging from diameters of less than 0.2 μm to a maximum of 15 μm. The average diameter and the number of globules are affected by endogenous, physiological, and exogenous factors. The size of the globules in turn affects the qualitative characteristics of milk and cheese. In addition, the average diameter of the globules and their surface that is exposed to the digestive system affect fat digestibility in different ways. Finally, the components of the MFG membranes have been shown to take part in the secretion process of globules and to have a beneficial effect on human health. In conclusion, by acting on factors influencing the dimensions of the fat globules and by increasing the content of the milk membrane could help adapt milk production to specific consumer targets and improve milk nutritional properties.

Cattle-yaks are increasingly cultivated to boost yak dairy production while reports claim that will decrease the nutritional value of yak milk. This study explores the therapeutic potential and mechanistic basis of a novel dairy-egg derived nutritional intervention-combining milk fat globule-epidermal growth factor 8 (MFG-E8) and egg yolk phosphatidylcholine (PC)-against dexamethasone (Dex)-induced skeletal muscle atrophy in aged rats. Specifically, we aimed to determine whether MFG-E8 + PC co-supplementation could mitigate muscle atrophy, improve mitochondrial function and elucidate the underlying mechanisms involving energy metabolism. By integrating targeted energy metabolomics, we demonstrate that the synergistic action of MFG-E8 and PC alleviates muscle atrophy in aged rats by restoring mitochondrial function and energy metabolism.

Aging is associated with aortic stiffening (AoSt), a condition characterized by diminished aortic elasticity that predisposes individuals to cognitive decline, including Alzheimer's disease (AD). Emerging evidence implicates medin, which is derived from milk fat globule-EGF factor 8 protein (MFG-E8), as a key link between AoSt and AD. Medin aggregates into aortic medial amyloid (AMA), which is found in approximately 97% of Caucasian individuals aged 50 and above, contributing to vascular inflammation, calcification, and loss of arterial elasticity.
