While coarse-grained (CG) simulations provide an efficient approach to identify small- and large-scale motions important to protein conformational transitions, coupling with appropriate experimental validation is essential. Here, by comparing small-angle X-ray scattering (SAXS) predictions from CG simulation ensembles of adenylate kinase (AK) with a range of energetic parameters, we demonstrate that AK is flexible in solution in the absence of ligand and that a small population of the closed form exists without ligand. In addition, by analyzing variation of scattering patterns within CG simulation ensembles, we reveal that rigid-body motion of the LID domain corresponds to a dominant scattering feature. Thus, we have developed a novel approach for three-dimensional structural interpretation of SAXS data. Finally, we demonstrate that the agreement between predicted and experimental SAXS can be improved by increasing the simulation temperature or by computationally mutating selected residues to glycine, both of which perturb LID rigid-body flexibility.

Purpose: Develop a musculoskeletal-environment interaction model to reconstruct the dynamic-interaction process in skiing.
Methods: This study established a skier-ski-snow interaction (SSSI) model that integrated a 3D full-body musculoskeletal model, a flexible ski model, a ski boot model, a ski-snow contact model, and an air resistance model. An experimental method was developed to collect kinematic and kinetic data using IMUs, GPS, and plantar pressure measurement insoles, which were cost-effective and capable of capturing motion in large-scale field conditions.

Functional imaging using genetically encoded indicators, such as GCaMP, has become a foundational tool for in vivo experiments and allows for the analysis of cellular dynamics, sensory processing, and cellular communication. However, large scale or complex functional imaging experiments pose analytical challenges. The increasing utilization of hard-to-cut materials in high-performance sectors such as aerospace and defense has pushed manufacturing systems to be flexible in processing large workpieces with a wide range of materials while also delivering high precision. Recent studies have highlighted the potential of integrating industrial robots (IRs) with electric discharge machining (EDM) to create a non-contact, low-force manufacturing platform, particularly suited for the accurate machining of hard-to-cut materials into complex and large-scale monolithic components. In response to this potential, a novel robotic EDM system has been developed.
