Although the ability of zinc to retard the oxidative process has been recognized for many years, zinc itself has been reported to induce oxidative stress. In order to give some insights into elucidating the role of intracellular Zn(2+) in cells suffering from oxidative stress, the effects of N-ethylmaleimide (NEM) and ZnCl(2) on cellular thiol content and intracellular Zn(2+) concentration were studied by use of 5-chloromethylfluorescein diacetate (5-CMF-DA) and FluoZin-3 pentaacetoxymethyl ester (FluoZin-3-AM) in rat thymocytes. The treatment of cells with NEM attenuated 5-CMF fluorescence and augmented FluoZin-3 fluorescence in a dose-dependent manner. These NEM-induced phenomena were observed under external Zn(2+)-free conditions. Results suggest that NEM decreases cellular thiol content and induces intracellular Zn(2+) release. Micromolar ZnCl(2) dose-dependently augmented both FluoZin-3 and 5-CMF fluorescences, suggesting that the elevation of intracellular Zn(2+) concentration increases cellular thiol content. Taken together, it is hypothesized that intracellular Zn(2+) release during oxidative stress is a trigger to restore cellular thiol content that is decreased by oxidative stress.

Ciprofloxacin (CFX) is a broad-spectrum antibiotic belonging to the fluoroquinolone class, widely used to treat bacterial infections by inhibiting bacterial DNA replication. Ferroptosis, a form of regulated cell death, is characterized by lipid peroxidation on cellular and organelle membranes. Our previous studies demonstrated that ciprofloxacin inhibits erastin-induced ferroptosis by enhancing glutathione peroxidase 4 (GPX4) protein stability.

Labile Zn is emerging as a quantitative driver, not just a biomarker, of metastasis, yet rapid, second-resolved intracellular measurement remains elusive. Here we engineer terpyridine-functionalised, hollow Au@SiO nanocapsules (NCs@TPY) and couple their SERS signal to cell-specific partial-least-squares (PLS) chemometrics, yielding an 8-log dynamic range (10 - 10 M), a low-nanomolar detection limit and ≤4.5 % cross-validated error while rejecting Ca/Mg interference.

3,4-dihydroxybenzenesulfonyl-functionalized polyethyleneimine (PS), a novel polymeric chelator, was synthesized by conjugating 3,4-dihydroxybenzenesulfonyl (CAM) groups with branched polyethyleneimine (BPEI, MW = 600 Da) via N-acylation. PS demonstrated a high uranium adsorption capacity of 78.08% at a concentration of 4 mg/mL, accompanied by significant selectivity over competing ions such as Ca, Zn, and Cu.

Intracellular zinc homeostasis and subcellular compartmentalization are exquisitely regulated, playing critical roles in immune regulation, such as inducing inflammatory programmed cell death, producing high levels of interferons and inflammatory cytokines, controlling the polarization and function of macrophages, . Herein, we designed two cyclometalated Ir(III) complexes with moderate zinc ion (Zn) affinity, capable of re-distributing the endogenous Zn from the cytoplasm and vesicles to mitochondria as indicated by ICP-MS measurement, thus inducing mitochondrial dysfunction and apoptosis. Moreover, mitochondria-targeted Ir2 also displayed better immunoregulation activity than lysosome-targeted Ir1, capable of triggering GSDMD-mediated pyroptosis caspase-1 dependent pathway and down-regulating PD-L1 levels in cancer cells.
