The vestibular cell type affected by congenital hypothyroidism (CH) was investigated by measuring the activity of glutamate decarboxylase (GAD) and choline acetyltransferase (ChAT), synthesizing enzymes of putative afferent (GABA) and efferent (acetylcholine, ACh) neurotransmitters and thus, respectively, hair cell I and II (HC-I, HC-II), and efferent terminal (ET) marker enzymes, in vestibular homogenates of control, congenitally hypothyroid rats (CHR) and in thyroxine-replaced CHR (CHR-T4) whose postnatal age ranged from 20 to 60 days old. In the vestibule, CH-II and its efferent cholinergic contacting bouton mature prior to thyroid function whereas HC-I-differentiation and its efferent synapse arrival are the latest events in vestibular maturation. Therefore, a differential effect of CH upon GAD and ChAT in CHR could be anticipated. In control rats as in CHR the magnitude of GAD was the same with time starting on the 20th day. In CHR, ChAT gradually diminished beginning on day 28 to become 45% decreased with respect to control on the 60th postnatal day. Prevention of ChAT decrease in CHR by early administration of thyroxine (T4), a striking diminution of T4 and triiodothyronine (T3) in CHR serum and a normal level of these hormones found in CHR-T4 corroborated thyroid involvement. These results confirm the preference of hypothyroidism to affect cholinergic cell types (or compartments) of late maturation (HC-I-containing ET and hence 45% ChAT decrease) leaving HC-I, HC-II and HC-II-connecting ET untouched, supported by a 55% remanent ChAT and a constant GAD activity regardless of time and treatment.

The present study provides new insights into the growth of the brown algal cell wall by showing that cell wall polysaccharides play an important role in the process of growth, considering the physicochemical characteristic of young and old . To determine its structural variation in detail, the cell wall was sequentially fractionated into five fractions: hot water (HW), ammonium oxalate, hemicellulose-I (HC-I), HC-II, and cellulose, and analyzed physicochemically. Results showed that almost 80% of the total recovery cell wall from both young and old thalli was HW, and HC-I contained mainly fucoidan composed of Fucose, Glucuronic acid, and sulfate in molar ratios of 1.

Brown algae contain a polysaccharide-rich cell wall, mainly composed of alginate and fucoidan which have been extensively studied for their individual structure and bioactivities. Particularly, the cell wall of Cladosiphon okamuranus is rich in fucoidan rather than alginate. However, little is known about its arrangement or interlinking with other polysaccharides such as cellulose in the cell wall.

Changes in pectin, hemicelluloses and cellulose in the cell walls of outer pericarp tissues of kiwifruit (Actinidia deliciosa cv. Hayward) were determined during development. An extensive amylase digestion was employed to remove possible contaminating starch before and after fractionation of wall polysaccharides.

The aim of the present study was to clarify the influence of obesity on the functions of low-density lipoprotein receptors (LDL-R) and 3-hydroxy-3-methylglutarate-coenyzme A (HMG-CoA) reductase both in healthy control subjects and in patients with hypercholesterolemia (HC). Experiments were performed on monocytes of 15 non-obese (C I) and 11 obese (C II) healthy control subjects and on 22 non-obese (HC I) and 26 obese (HC II) patients with HC. [(125)I]LDL was used to determine LDL-R activity by measuring binding and intracellular degradation.
