Dysregulation of the RAF-MEK-ERK1/2 pathway is involved in the pathoetiology of many diseases. Its central role in cancer has led to the development of drugs targeting upstream receptors, RAS, and kinases in the extracellular signal-regulated kinase 1 (ERK1) and 2 (ERK2) signaling cascade. The use of these drugs in cancer therapy - together with ongoing monitoring of their effectiveness, evolving side-effects, and resistance mechanisms - has expanded our knowledge of both the physiological and pathological functions of ERK1/2 and could thus provide potential alternative therapeutic strategies.

The aim of this study was to investigate three unrelated Simmental calves with atypical white coat color, identify potential genetic causes using a trio-based whole-genome sequencing approach, and assess the prevalence of the identified variants in the breed. Several inherited alleles affecting coat color, ranging from fawn to red spotted and white-headed, have been described in Simmental cattle originating from Switzerland. However, no genetic variant has yet been associated with an almost completely white coat in this breed.

Left atrial appendage occlusion (LAAO) is an alternative to oral anticoagulation for stroke prevention in atrial fibrillation (AF), especially in patients at high bleeding risk. While generally safe, LAAO involves procedural and postprocedural risks. The most serious complication is pericardial effusion or tamponade; other periprocedural risks include stroke, device embolization, vascular injury, major bleeding, and air embolism.

Objective: Evaluate the audiological outcomes of the Cochlear Osia 2 Bone Conduction System implanted in children ages 5-11 years old.

Design: A pivotal, prospective, open-label, multicentre clinical trial to expand access to the Osia 2 system to children ages 5-11 years old.

Study Sample: Children aged 5-11 years old who presented with either (1) a conductive or mixed hearing loss where the pure tone average bone conduction threshold (measured at 500, 1000, 2000, and 3000 Hz) was ≤ 55 dB HL or (2) single-sided deafness where the ear to be implanted had a profound sensorineural hearing loss and the pure tone average air conduction threshold (measured at 500, 1000, 2000, and 3000 Hz) in the contralateral ear was ≤ 20 dB HL.

The targeted and personalized cell and gene therapy with autologous chimeric antigen receptor (CAR) transduced T‑cells (CAR T‑cells) has become established as the standard therapy for patients with B‑cell diseases, such as CD-19 positive leukemia and lymphomas and B‑cell maturation antigen (BCMA) positive multiple myeloma (MM). This therapy has been approved in Europe since 2018 and is administered in more than 50 clinics in Germany. With increasing experience and expertise in understanding of the principles of action and underlying mechanisms as well as mitigation of side effects have significantly improved.
