Mastocytosis is a heterogeneous group of hematopoietic disorders characterized by abnormal growth and accumulation of mast cells (MC) in one or more organs. Clinical symptoms occur as a result of the release of chemical mediators and/or of pathologic infiltration of MC in various tissues. Although the initial events leading to mastocytosis have not yet been unraveled, acquired alterations in the c-kit gene coding for the receptor of stem cell factor (SCF), a major cytokine involved in MC growth, have been described in a significant number of patients. Of particular interest are point mutations resulting in a constitutively activated SCF receptor. Such mutations are probably involved in the abnormal (SCF-independent) proliferation of MC in these patients. New therapeutic strategies may be envisaged to inhibit the deregulated kinase activity of these mutant forms of c-kit.

Food combinations featuring specific functional components represent one of the effective intervention strategies for alleviating functional gastrointestinal disorders induced by dietary and environmental factors. Honey and aloe vera have both been recognized as natural agents with laxative effects, yet the synergistic effects of their combination in alleviating constipation and the underlying regulatory mechanism remain to be elucidated. This study formulated a honey-aloe paste by employing honey as the primary ingredient compounded with aloe vera gel and investigated its preventive effects on loperamide-induced slow-transit constipation through a comprehensive analysis of gastrointestinal function and intestinal microenvironment.

Purpose: To evaluate the efficacy and underlying mechanism of advanced optimal pulse technology intense pulsed light (AOPT) in low-energy triple-pulse long-width mode (AOPT-LTL) for melasma treatment.
Methods: An in vivo guinea pig model of melasma was established through progesterone injection and ultraviolet B radiation. Three sessions of AOPT-LTL treatment were performed weekly.

Overexpression and gain-of-function mutations of c-Kit have been implicated in cancers including gastrointestinal stromal tumors (GIST), small cell lung cancer (SCLC), acute myeloid leukemia, and systematic mastocytosis. In clinics, small molecule c-Kit inhibitors often result in secondary c-Kit mutations or are ineffective despite c-Kit overexpression. We developed NN3201, a novel c-Kit targeting antibody-drug conjugate (ADC), via rational design to evaluate its anticancer activity in c-Kit-positive tumors, and preclinical pharmacologic profiles.
