Infantile epileptic spasms syndrome (IESS) is a rare, early-onset pediatric epilepsy with severe neurological consequences. In refractory cases, the ketogenic diet (KD) is used as a metabolic therapy. To understand the antiepileptic mechanisms of the diet, this study examined the proteomic profiles in the hippocampus after KD treatment.

Mitochondrial respirometry provides a detailed assessment of oxygen consumption within the electron transport system, yet methods detailing respiration from non-invasive samples remain limited. Here, we present a protocol for measuring mitochondrial respiration in cultured buccal cells. We outline procedures for buccal cell collection, primary cell culture, and respirometry calibration, followed by oxygen consumption measurements and cell count for data normalization.

L-Lactate is increasingly appreciated as a key metabolite and signaling molecule in mammals. However, investigations of the inter- and intra-cellular dynamics of L-lactate are currently hampered by the limited selection and performance of L-lactate-specific genetically encoded biosensors. Here we now report a spectrally and functionally orthogonal pair of high-performance genetically encoded biosensors: a green fluorescent extracellular L-lactate biosensor, designated eLACCO2.
