Background Non-clear cell renal cell carcinomas (nccRCCs) are a histologically heterogeneous and clinically underrepresented group of renal malignancies, distinct from clear cell RCC in morphology, molecular profile, and prognosis. Despite accounting for 20-30% of RCCs, nccRCC subtypes are infrequently studied, especially in the Indian population. We recently showed that vascular normalization using anti-VEGF therapy can overcome these challenges and improve the outcome of CAR-T therapy in glioblastoma models in mice. Here, we developed a physiologically based pharmacokinetic model to simulate the dynamics of both adoptively transferred CAR-T cells and endogenous immune cells in solid tumors following vascular normalization.

Purpose: Genetic variation underlying rare diseases in Arab populations is poorly understood limiting effective carrier screening for recessive disorders, which are prevalent because of high consanguineous rates.
Methods: Using the American College of Medical Genetics and Genomics/Association for Molecular Pathology guidelines, we curated pathogenic (P) and likely pathogenic (LP) variants in 1333 Arab Emirati families (346 internal cohort and 987 from the literature). We also analyzed P/LP variants in 1194 Emirati exomes, calculated allele frequencies, and estimated carrier rates for the associated recessive conditions.
Opioid use disrupts the hypothalamic-pituitary-adrenal (HPA) axis, altering stress responses depending on the stage of use. A higher cortisol/DHEA-S ratio may indicate relapse risk in substance use disorders. This study aimed to explore differences in serum cortisol/DHEA-S ratio and its association with proximal and perceived stress, social support, and coping strategies in opioid-dependent youth.
