In ephaptic coupling, physically adjacent neurons influence one another's activity via the electric fields they generate. To date, the molecular mechanisms that mediate and modulate ephaptic coupling's effects remain poorly understood. Here, we show that the hyperpolarization-activated cyclic nucleotide-gated (HCN) channel lateralizes the potentially mutual ephaptic inhibition between gustatory receptor neurons (GRNs).

Drosophila TrpA1 (transient receptor potential ankyrin 1) transcripts are alternatively spliced at 2 distinct sites each with a choice of mutually exclusive exons. The first site determines exon1 encoding the amino terminus to produce either nucleophile-, electrophile- and noxious temperature-gated TRPA1(A) or electrophile- and innocuous warmth-gated TRPA1(B). The second site selects for exon10, resulting in TrpA1 variants with either exon10a or exon10b encoding a domain between the N-terminal ankyrin repeats and the transmembrane segments.

Establishing transepithelial ion disparities is crucial for sensory functions in animals. In insect sensory organs called sensilla, a transepithelial potential, known as the sensillum potential (SP), arises through active ion transport across accessory cells, sensitizing receptor neurons such as mechanoreceptors and chemoreceptors. Because multiple receptor neurons are often co-housed in a sensillum and share SP, niche-prevalent overstimulation of single sensory neurons can compromise neighboring receptors by depleting SP.

Article Synopsis
  • - The study focused on the effects of mammographic breast density reduction (MDR) on treatment outcomes in premenopausal women with estrogen receptor-positive breast cancer who received either tamoxifen alone or in combination with ovarian function suppression (OFS).
  • - Results from the ASTRRA trial showed that while the rate of MDR positivity was similar in both treatment groups, patients receiving tamoxifen plus OFS who were MDR-positive experienced significantly better disease-free survival compared to those who were MDR-negative.
  • - The findings highlight the potential importance of MDR as a positive prognostic factor specifically in patients treated with tamoxifen and OFS, suggesting a need for tailored treatment approaches based on MDR status.

Purpose: Fertility preservation (FP) is an important issue for young survivors of breast cancer. Although international guidelines recommend pre-treatment fertility counseling for women with breast cancer, there is no standardized protocol or referral system for FP in South Korea. There are also barriers to discussing FP that make patient-centered decision making difficult.

Article Synopsis
  • Scientists are studying special gene changes (called PTVs) to see how they affect breast cancer in 8,852 Asian patients.
  • They found that having these gene changes is linked to more aggressive tumors, which are harder to treat.
  • Overall survival rates for patients with these gene changes were lower, suggesting that some genes might play a role in how long patients live after being diagnosed with breast cancer.

Background: Genetic testing for breast cancer susceptibility is widely used, but for many genes, evidence of an association with breast cancer is weak, underlying risk estimates are imprecise, and reliable subtype-specific risk estimates are lacking.

Methods: We used a panel of 34 putative susceptibility genes to perform sequencing on samples from 60,466 women with breast cancer and 53,461 controls. In separate analyses for protein-truncating variants and rare missense variants in these genes, we estimated odds ratios for breast cancer overall and tumor subtypes.
