Anthraquinones are the major bioactive compounds in alder buckthorn, a plant with a long history of use for the treatment of obstipation in European and Asian traditional medicine. As these metabolites are also associated with toxic side effects, reliable analytical techniques for their determination are essential. Previously described methods mainly utilized Liquid Chromatography for this purpose, as the photometric assay described in the current edition of the European Pharmacopoeia lacks selectivity.

Article Synopsis
  • * A supercritical fluid chromatography technique was developed to simultaneously identify and quantify seven alkaloids in less than six minutes, showing high precision and accuracy per ICH guidelines.
  • * The study also applied this method to isolate five specific alkaloids from a crude extract using centrifugal partition chromatography, achieving good yields and confirming their purity with further analysis.

()-3-(4-(()-1-(4-Hydroxyphenyl)-2-phenylbut-1-enyl)phenyl)acrylic acid () as a carrier was esterified with alkenols of various lengths and coordinated through the ethylene moiety to PtCl, similar to Zeise's salt (K[PtCl(CH)]). The resulting complexes (Alk = Prop, But, Pent, Hex) degraded in aqueous solution only by exchange of the chlorido ligands. For example, coordinated the amino acid alanine in the cell culture medium, bound the isolated nucleotide 5'-GMP, and interacted with the DNA (empty plasmid pSport1).

Different Senecio species, especially S. inaequidens - a neophyte native to South Africa - have widely spread across Europe and now are found worldwide. The entire genus is known to contain toxic pyrrolizidine alkaloids (PAs), which renders them a possible health hazard to humans and livestock.

Article Synopsis
  • * A new ultrahigh-performance liquid chromatography (UHPLC) method was developed to efficiently separate and analyze eleven different MAAs from various algal species in under 8 minutes.
  • * The method was validated for precision and accuracy, showing consistent results across different algae, highlighting significant variation in MAA profiles and concentrations, particularly for the common MAA shinorine.

Marine rhodophyta are known to synthesize specific secondary metabolites, mycosporine-like amino acids (MAAs), to protect themselves from harmful UV-radiation. Shinorine and porphyra-334 are among the most abundant representatives of this compound class. In the present work, a novel approach for their isolation is described.

As recently shown, some fungal pigments exhibit significant photoactivity turning them into promising agents for the photodynamic treatment of microbial infections or malignant diseases. In the present study, a separation strategy for fungal anthraquinones was developed based on centrifugal partition chromatography. A suitable method was explored employing a methanolic extract of the fruiting bodies of Cortinarius sanguineus (Agaricales, Basidiomycota).

Boswellic acids, a class of triterpenes, are the bioactive constituents in Indian frankincense, an herbal drug with pronounced anti-inflammatory activity. In this study their separation and quantification in B. serrata extracts is reported for the first time by using Supercritical Fluid Chromatography.

HLA-DPB1 antigens are mismatched in about 80% of allogeneic hematopoietic stem cell transplantations from HLA 10/10 matched unrelated donors and were shown to be associated with a decreased risk of leukemia relapse. We recently developed a reliable in vitro method to generate HLA-DPB1 mismatch-reactive CD4 T-cell clones from allogeneic donors. Here, we isolated HLA-DPB1 specific T cell receptors (TCR DP) and used them either as wild-type or genetically optimized receptors to analyze in detail the reactivity of transduced CD4 and CD8 T cells toward primary AML blasts.
