Purpose: Systemic treatment options for vulvar squamous cell carcinoma (VSCC) are limited. ROS1, a tyrosine kinase implicated, for example, in non-small cell lung cancer (NSCLC), has recently shown responsiveness to tyrosine kinase inhibitors. This study investigated immunohistochemical ROS1 expression in VSCC to explore its potential as a future therapeutic target in this rare malignancy.

In the present study, we evaluated 68 patients with primary vulvar carcinomas (VC) and analyzed the cytoplasmic/membranous immunohistochemical expression of podoplanin in viable tumor cells and cancer-associated fibroblasts (CAFs) within the adjacent peritumoral stroma. To determine potential associations between podoplanin expression and traditional clinicopathological parameters (e.

In the present study, we therefore establish a supervised machine learning algorithm to predict groin metastasis in patients with squamous cell carcinoma of the vulva (VSCC) based on classical histomorphological features. : In total, 157 patients with VSCC were included in this retrospective study.

However, the prognostic relevance of morphological biomarkers such as tumor budding (TB) and poorly defined clusters (PDCs) remains to be determined. We histologically analyzed the formation of peritumoral and intratumoral TB and PDCs in a cohort of 157 patients with VC.

Background: The prognostic relevance of tumor-infiltrating lymphocytes (TILs) has so far been recognized in several solid tumors like in breast, endometrial and ovarian cancer-nonetheless, the immune contexture of squamous cell carcinomas of the vulva, analyzed by means of stromal (s) and intratumoral (i) TILs, remains yet to be elucidated.
Material And Methods: In this study, we examined the immunooncological biomarkers sTILs and iTILs in 157 vulvectomy specimens with histologically diagnosed vulvar squamous cell carcinoma (VSCC) according to the standardized methodology proposed by the International Immunooncology Biomarkers Working Group in 2017. In a next step, we evaluated the association of infiltrating lymphocytes to traditional histopathological parameters such as infiltration depth and HPV related tumorigenesis.
: To evaluate the prognostic impact of immunohistochemical ki-67 staining analysis regarding lymph node involvement and survival data (overall/progression-free survival) in squamous cell carcinoma of the vulva. : A systematic literature search of English and German articles was conducted (PubMed, Embase, Scopus, Web of Science) from 1980 to December 2023, including the search terms "vulvar Neoplasms", "vulvar cancer", "vulvar carcinoma", "vulvar tumor", "vulvar tumour", "vulvar malignancy", "vulvar malignant", "ki-67", "MIB-1", "MIB1", "proliferative index", "proliferative activity", "mitotic index", and "mitotic count". Study quality was assessed using a two-step "mixed-criteria" approach; to synthesize study results, a narrative summary is provided.

Current prognostic biomarkers are often costly and methodologically demanding. In this regard, histomorphological biomarkers such as tumor buds (TB) and poorly differentiated clusters (PDC) represent promising, cost-effective prognostic indicators that are relatively straightforward to implement.

Stromal-epithelial interaction is thought to have a major impact on cancer development and progression. Previous studies have shown that interaction via soluble factors lead to a reduction in the expression of xCT and AL122023.

In most cases, multimodal therapeutic approaches are required and show only limited cure rates with a high risk of tumor recurrence. Anti-PD-1 antibody treatment was recently approved for recurrent and metastatic cases but to date, response rates remain lower than 25%.

Research in human tissue is insufficient. Our aim was to investigate the distribution of TRPC1, 3, and 6 in the healthy human thyroid.

From an oncological perspective, however, immunohistochemical analysis of claudin 18.2 protein expression has become increasingly clinically relevant nowadays since the impressive therapeutic benefits of the claudin 18.

Consecutively, the present study examines the immunohistochemical expression of the calcium transient receptor potential channels 3 and 6 (TRPC3 and TRPC6) in ectopically located (outside the uterine cavity) endometrial tissue.
Methods: Laparoscopically collected and histomorphologically verified endometriosis tissues from several different intraabdominal locations were examined (n = 20) and immunohistochemical stainings were performed with anti-TRPC3 and anti-TRPC6 antibodies (Alomone Labs, Jerusalem).
Background: Histopathological biomarkers of carcinomas and their prognostic relevance, such as Broder's grading system (based on the total number of undifferentiated cells) or Bryne's grading system (rating morphological features at the tumor invasive front), have been repeatedly and successfully put to test. Since most studies focus on head and neck cancers or oral carcinomas, for squamous cell carcinoma of the vulva, no standardized and agreed on pathological tumor grading system, yielding prognostic significance, could be determined so far.
Material And Methods: To determine prognostic associations of different grading systems with regard to groin lymph node metastasis, 73 cases of vulvar carcinomas (VC) were re-examined within our study and Broder's and Bryne's grading system individually performed.

Early detection of PDAC remains challenging due to the lack of early symptoms and the absence of reliable biomarkers. The aim of the present project was to identify miRNA and proteomics signatures discriminating PDAC patients with DM from nondiabetic PDAC patients. Proteomics analysis and miRNA array were used for protein and miRNA screening.

While TRPC6 subtype detection in submandibular glands and the relevance of some TRPC channels in this gland have been shown in animal models, its histological detection in human lacrimal and submandibular glands, as well as related tumors, lacks systematic study. Studying TRPC6 in humans could lead to new therapeutic options.

Despite a proposed impact of nationwide lockdowns in response to the COVID-19 pandemic on oncological diseases, the effect on VC staging and tumor characteristics remains yet to be resolved; therefore, analyzing pathological data from patients with squamous cell VC pre-, during, and post-COVID in a high-incidence region may offer insights into potential epidemiological and clinical trends. : We identified a total of 90 patients who were diagnosed at the Institute of Pathology, University Hospital Saarland, between 2018 and 2023, and defined three distinct cohorts: a pre-COVID cohort (2018-2019), a COVID cohort (2020-2021), and a post-COVID cohort (2022-2023).
