Known for its widespread outbreaks, including the 2013-2016 epidemic that infected almost 29 000 individuals and resulted in approximately 11 300 deaths, Ebola virus (EBOV) and related filoviruses remain a current threat as consecutive filoviral outbreaks have occurred between 2021 through 2025. Due to high fatality rates of 40-90% among infected individuals, researchers have invested significant efforts to discover effective treatments for Ebola virus disease. Small molecules hold great potential for treating Ebola virus disease because they can target various stages of the filoviral life cycle, such as entry, transcription, replication, and egress; however, the FDA has not yet approved any small molecule treatments for EBOV.

Background: Recruitment to cancer clinical trials (CCTs) is low, particularly for underrepresented groups such as uninsured patients, those with low-income status, and racial and ethnic minoritized individuals. A significant barrier is that treating oncologists often fail to inform patients about the possibility of CCT participation as an option for quality cancer care. Therefore, patient inquiries about trials before starting treatment should be normalized and encouraged, particularly for underrepresented groups.

Saturn is characterized by strong zonal winds at the cloud level, with a wide equatorial west-to-east flow, two retrograde jets on its flanks, and a series of mid-to-high latitude jets in both hemispheres. Based on the Cassini mission gravity measurement up to zonal harmonic J, it was shown that these winds penetrate to approximately 9000 km. However, these analyses relied fully on the low, even harmonics dominated by an interior structure that is not known to high accuracy.

Io experiences tidal deformation as a result of its eccentric orbit around Jupiter, which provides a primary energy source for Io's continuing volcanic activity and infrared emission. The amount of tidal energy dissipated within Io is enormous and has been suggested to support the large-scale melting of its interior and the formation of a global subsurface magma ocean. If Io has a shallow global magma ocean, its tidal deformation would be much larger than in the case of a more rigid, mostly solid interior.

Since the largest and most fatal Ebola virus epidemic during 2014-2016, there have been several consecutive filoviral outbreaks in recent years, including those in 2021, 2022, and 2023. Ongoing outbreak prevalence and limited FDA-approved filoviral therapeutics emphasize the need for novel small molecule treatments. Here, we showcase the structure-activity relationship development of N-substituted pyrrole-based heterocycles and their potent, submicromolar entry inhibition against diverse filoviruses in a target-based pseudovirus assay.
