Efficacy of Liposomal Bupivacaine in Ultrasound-Guided Peripheral Nerve Blocks for Postoperative Pain Management in Foot and Ankle Surgery.

Background: In response to the opioid epidemic, many surgical specialties have adopted nonopioid pain management strategies. Ultrasound (US)-guided peripheral nerve blocks (PNBs) are effective in reducing pain and opioid consumption postsurgery. Liposomal bupivacaine (LB), shown effective in shoulder surgery, was approved in November 2023 for use in US-guided lower extremity blocks.

ssDNA and ssRNA Promote Phase Condensation of SAMHD1.

SAMHD1 (SAM domain and HD domain-containing protein 1) is a deoxynucleoside triphosphate triphosphohydrolase (dNTPase) with functions in viral restriction, R-loop resolution, DNA repair, telomere maintenance, ssRNA homeostasis, and regulation of self-nucleic acids. As a dNTPase, SAMHD1 functions as an allosterically activated tetramer, where binding of GTP to the A1 activator site of each monomer initiates dNTP-dependent tetramerization. cEM structures reveal that the nucleic-acid-related functions of SAMHD1 involve binding of guanine residues to the A1 site, leading to oligomeric forms that appear as beads-on-a-string on single-stranded RNA and DNA.

Increase in alcohol outlets by neighbourhood socioeconomic status following the expansion of alcohol sales into convenience stores in Ontario, Canada.

Introduction: In September 2024, the province of Ontario expanded alcohol sales into convenience stores. It is important to examine differences in alcohol sales expansion by socioeconomic status (SES) as lower SES groups experience a disproportionately higher burden of alcohol-attributable harms.

Methods: This repeated cross-sectional study examined whether neighbourhood SES was associated with increases in alcohol outlets immediately following the expansion of alcohol sales.

Introduction: Stroke is the second leading cause of death worldwide, with the greatest burden in low- and middle-income countries (LMICs). Haemorrhagic stroke or spontaneous intracranial haemorrhage (sICH), including intraparenchymal haemorrhage (IPH) and subarachnoid haemorrhage (SAH), has the highest mortality and morbidity. Local management practices for haemorrhagic stroke vary greatly between geographical regions.
