Multi-Institutional Study of ALK-Positive Large B-Cell Lymphoma: Outcomes in the Era of ALK Inhibitors and Biologically Informed Therapies.

Anaplastic lymphoma kinase-positive large B-cell lymphoma (ALK+ LBCL) is a rare, aggressive subtype of diffuse large B-cell lymphoma with poor outcomes using standard chemotherapy. In this multi-institutional retrospective study, we analyzed 39 cases of ALK+ LBCL identified at six US academic centers from 2002 to 2024, with treatment including conventional cytotoxic regimens in frontline and biologically informed and nonchemotherapy-based strategies in the relapsed setting. Ninety-two percent of patients received frontline anthracycline-based chemotherapy; 43% received intensified regimens, and 15% underwent upfront autologous stem cell transplantation (ASCT).

Case Summary: A 27-year-old man with neurofibromatosis type 2 on long-term bevacizumab presented with progressively worsening chest pain and subsequently experienced ventricular fibrillation arrest.

Incorporating novel therapies into the treatment of older patients with classic Hodgkin lymphoma.

Introduction: Outcomes of older patients with classic Hodgkin lymphoma (cHL) are inferior to those of younger patients secondary to their distinct clinical presentation, unique disease biology, increased burden of medical comorbidity, and poorer tolerance to conventional therapies. Greater attention to the objective parameters underlying patient fitness has led to the recognition of comprehensive geriatric assessments (CGA) as an important method to optimize selection for appropriate therapy intensity. Given the magnified risk associated with traditional chemotherapy in older patients with cHL, the incorporation of the novel agents brentuximab vedotin (BV) and immune checkpoint inhibitors (ICIs) into modern treatment paradigms offers promise for improved outcomes in this population.

Avelumab and taxol chemotherapy in platinum-refractory or ineligible metastatic urothelial carcinoma (AVETAX trial).

Background And Objective: This study investigates the role of taxanes and immunotherapy for inoperable advanced or metastatic urothelial carcinoma in patients unable to tolerate or refractory to platinum-based regimens. We aimed to determine the safety and estimate the efficacy of a combination of avelumab and docetaxel in treating advanced or metastatic urothelial carcinoma.

Methods: This phase 1b, single-arm, open-label clinical trial involved adults with advanced or metastatic urothelial carcinoma, progressing during or after platinum-containing chemotherapy, within 12 months of neoadjuvant or adjuvant platinum-containing chemotherapy, or ineligible for platinum regimens.

Effect of peritransplant measurable residual disease clearance in patients with myelodysplastic neoplasm: a referral center experience.

Pre-allogeneic stem cell transplant (pre-HSCT) measurable residual disease (MRD) is increasingly recognized as a prognostic marker. However, the MRD status in myelodysplastic neoplasm (MDS) or myelodysplastic/myeloproliferative neoplasm (MDS/MPN) are less well-defined. We performed a retrospective chart review of adults who underwent HSCT for MDS or MDS/MPN in 2012-2023 and evaluated the effect of pre-HSCT MRD status on relapse-free survival (RFS) and overall survival (OS).

The communication between neural stem cells (NSCs) and surrounding astrocytes is essential for the homeostasis of the NSC niche. Intercellular mitochondrial transfer, a unique communication system that utilizes the formation of tunneling nanotubes for targeted mitochondrial transfer between donor and recipient cells, has recently been identified in a wide range of cell types. Intercellular mitochondrial transfer has also been observed between different types of cancer stem cells (CSCs) and their neighboring cells, including brain CSCs and astrocytes.

Venetoclax-based therapy in treatment-naïve and relapsed/refractory acute myeloid leukemia.

Neutropenic Enterocolitis: A Rare Complication of Sacituzumab Govitecan.

Neutropenic enterocolitis (NE) is a rare form of inflammatory colitis seen in severely neutropenic patients. Patients with NE typically have severe neutropenia (absolute neutrophil count [ANC] <500/mm) in the setting of exposure to cytotoxic chemotherapy. Taxanes have traditionally been associated with NE, but there has been a growing amount of literature, linking a variety of other chemotherapeutic agents.

Objective: Given the importance of scholarly work in academic medicine, better understanding of the manuscript review process (MRP) is useful for authors, reviewers, and editorial boards. We aim to describe the MRP at the Journal of Pediatric Gastroenterology and Nutrition (JPGN), assess the correlation between editor decisions and reviewer recommendations, and provide transparency to this process.

Methods: All manuscripts submitted in 2018 to JPGN were included in this analysis.

Herpes Simplex Virus 1 Infection Promotes the Growth of a Subpopulation of Tumor Cells in Three-Dimensional Uveal Melanoma Cultures.

Herpes simplex virus 1 (HSV-1)-mediated oncolytic therapy is an emerging cancer treatment modality with potential effectiveness against a variety of malignancies. To better understand the interaction of HSV-1 with neoplastic cells, we inoculated three-dimensional (3D) cultures of human uveal melanoma cells with HSV-1. 3D melanoma cultures were established by placing tumor cells on the surface of a Matrigel matrix, which was followed by the growth of tumor cells on the matrix surface and invasion of the Matrigel matrix by some tumor cells to form multicellular tumor spheroids within the matrix.

The impact of aging, hearing loss, and body weight on mouse hippocampal redox state, measured in brain slices using fluorescence imaging.

The relationships between oxidative stress in the hippocampus and other aging-related changes such as hearing loss, cortical thinning, or changes in body weight are not yet known. We measured the redox ratio in a number of neural structures in brain slices taken from young and aged mice. Hearing thresholds, body weight, and cortical thickness were also measured.
