Background: Lower extremity lymphedema (LEL) can develop in patients who undergo inguinal lymph node dissection (ILND) in the treatment of gynecologic, genitourinary, and skin and soft tissue malignancies. While LEL can negatively impact quality of life, the poorly documented prevalence and severity of lymphedema-related symptoms complicates the ability to identify high-risk patients and improve the selection of candidates for emerging microsurgical interventions.

Methods: This multicenter, cross-sectional study included patients who underwent ILND between 1990 and 2022 across three medical centers in the Netherlands.

Aim: We previously published selection criteria for proton therapy (PT) in the Netherlands for breast cancer (BC) and thoracic lymphoma patients, based upon the Absolute Excess Risk (AER) for acute coronary events. The aim of the current paper was to define additional criteria to select BC and thoracic lymphoma patients for PT, to reduce the risk of second primary BC and lung cancer (LC).

Methods: Based on a literature review, we identified Excess Relative Risks (ERR)/Gy mean organ dose of second primary BC and LC, for selected patient groups.

The multitude of DNA lesion types, and the nuclear dynamic context in which they occur, presents a challenge for genome integrity maintenance as this requires the engagement of different DNA repair pathways. Specific "repair controllers" that facilitate DNA repair pathway crosstalk between double-strand break (DSB) repair and base excision repair (BER) and that regulate BER protein engagement at lesion sites have yet to be identified. Here, we find that DNA polymerase β (Polβ), crucial for BER, is ubiquitylated in a BER complex-dependent manner by TRIP12, an E3 ligase that partners with UBR5 to restrain DSB repair signaling.

Purpose: Patritumab deruxtecan (HER3-DXd; MK-1022) is an investigational HER3-directed antibody-drug conjugate composed of a human immunoglobulin G1 monoclonal antibody to HER3 (patritumab) covalently linked via a stable tetrapeptide-based cleavable linker to a topoisomerase I inhibitor payload that has shown durable antitumor activity in previously treated patients with -mutated advanced non-small cell lung cancer (NSCLC). We extend these observations to patients with advanced NSCLC with other/no identified driver genomic alterations.

Methods: Patients with advanced squamous or nonsquamous NSCLC without a common -activating mutation whose disease had progressed on previous therapies including platinum-based chemotherapy, immune checkpoint inhibitors, and targeted therapy (for patients with known actionable genomic alterations) received HER3-DXd 5.
