The integration of wearable medical devices and digital health technologies (DHTs) in health care has grown significantly during the past 2 decades, particularly in dermatology, in which objective measurement of symptoms such as itch remains challenging. This review examines the evolution of DHTs in dermatology, focusing on the validation frameworks necessary for their implementation in clinical trials and research. We discuss the key stages of validation: hardware validation to ensure device reliability, analytical validation to transform raw sensor data into meaningful metrics, and clinical validation to demonstrate utility in specific patient populations.

Epithelial tissues are populated with accessory cells such as immune cells, sensory cells, and pigment-producing melanocytes, which must migrate through and intercalate between tightly adherent epithelial cells. Although much is known about how cells migrate through interstitial spaces consisting of predominantly of collagen-rich ECM and mesenchyme, how cells migrate through confined epithelial spaces without impairing barrier function is far less understood. Here, using live imaging of the mouse epidermis, we captured the migration of embryonic melanocytes (melanoblasts) while simultaneously visualizing the basement membrane or epithelial surfaces.

Obstructive defecation syndrome (ODS) is a condition that causes straining and may require manual evacuation and patients' thoughts circle around defecation impairing their quality of life. Mental comorbidity-related findings suggest a mental burden in ODS patients. In an observational cohort design, this study investigated the relationship between the mental distress and their clinical symptom severity for a group of patients with ODS who underwent surgical treatment.

Graph-based machine learning methods are valuable tools for identifying and predicting variation in genetic data. In particular, understanding phenotypic effects at the cellular level is an accelerating area in pharmacogenomics. Insight into how drugs or disease affect bio-networks could aid drug development and precision medicine.
