DNA sequencing using nanopores is now well established. Could the approach be extended to proteins ? There are a number of difficulties but work by many groups has shown proof-of-principle solutions for most of them. Further progress could indeed lead to a workable protein sequencing approach that would have many applications in research and in the clinic.

Motivation: Studying protein isoforms is an essential step in biomedical research; at present, the main approach for analyzing proteins is via bottom-up mass spectrometry proteomics, which return peptide identifications, that are indirectly used to infer the presence of protein isoforms. However, the detection and quantification processes are noisy; in particular, peptides may be erroneously detected, and most peptides, known as shared peptides, are associated to multiple protein isoforms. As a consequence, studying individual protein isoforms is challenging, and inferred protein results are often abstracted to the gene-level or to groups of protein isoforms.

For the last decade, the field of DNA sequencing has been dominated by a single company, Illumina, with very limited competition. However, a new method with superior performance and lower cost has recently been announced by Roche, a major pharmaceutical and diagnostics company, and promises to have a large impact, especially (but not exclusively) for medical applications.

For families of children with medical complexity (CMC), lack of nursing support in the home can result in significant social and financial hardships beyond its effects on clinical outcomes. This study aimed to quantify the impact of limited availability of private duty nursing. Families of CMC reported that inadequate access to in-home nursing creates significant downstream effects including delays in hospital discharge, major career changes among parents, especially on mothers, and significant annual income loss.
